SUMMARY:Graduate Student Seminar Series - Saleh Tabatabaei
DESCRIPTION:Graduate Student Seminar Series\nPlease ensure you invite your Principal Investigator by adding their email via the ‘Add Guest’ button and they will also be notified of your presentation.\nLocation: MS2158 – 1 King’s College Circle\nPresentation Title: Quantification of 3D Musculoskeletal CT Image-Based Biomarkers in the Lumbar Spine: Application in Metastatic Prostate Cancer\nAbstract:\nOsteosarcopenia\, characterized by concurrent deterioration of bone and muscle\, is increasingly recognized as a major contributor to frailty\, fractures\, and mortality\, particularly in prostate cancer. However\, quantitative longitudinal assessment of osteosarcopenia remains limited by existing manual methods based on 2D analysis at a single time point\, which are impractical for large-scale monitoring and cannot detect subtle changes over time due to slice-selection variability.\nThis study introduces an AI-enabled pipeline to quantify musculoskeletal (MSK) changes from 3D serial CT scans in prostate cancer. The pipeline integrates deep learning–based vertebrae detection and 3D vertebral body and psoas muscle segmentation. Lumbar vertebral bone density\, psoas muscle volume and density were retrospectively quantified from 5\,966 CT scans acquired from a cohort of 916 metastatic prostate cancer patients (2008–2024).\nThe pipeline demonstrated strong agreement with a manually contoured test set for vertebral density (R^2=0.874-0.986)\, psoas density (R^2=0.955)\, and psoas volume (R^2=0.830). In repeated short-interval scans\, the proposed 3D biomarkers showed higher reliability than conventional 2D measurements for psoas density (ICC(3\,1)=0.86 vs. 0.72) and psoas volume versus cross-sectional area (ICC(3\,1)=0.94 vs. 0.87). Longitudinal mixed-effects modeling showed that declining muscle and bone biomarkers were associated with increasing lumbar metastatic burden (p<0.05). The combined muscle model explained the greatest proportion of variance (marginal R^2=0.424).\nThese findings demonstrate that automated 3D CT-based biomarkers provide reliable and clinically meaningful measures of progressive MSK decline. This framework enables scalable opportunistic monitoring of osteosarcopenia and may support earlier identification of patients at risk for treatment-related deterioration\, metastatic progression\, and fracture.\nSupervisor Name: Dr. Cari Whyne / Dr. Michael Hardisty\nYear of Study: 5\nProgram of Study: PhD\nPowered by Calendly.com

DESCRIPTION:Graduate Student Seminar Series\nPlease ensure you invite your Principal Investigator by adding their email via the ‘Add Guest’ button and they will also be notified of your presentation.\nLocation: MS2158 – 1 King’s College Circle\nPresentation Title: 3D printable single ventricle model for cardiac tissue engineering\nAbstract: Engineering functional cardiac tissues requires biomaterials that replicate the structural\, mechanical\, and electrical properties of the native myocardium. Here\, we developed a 3D-printable\, porous\, and electrically conductive biomaterial for fabricating a single-ventricle cardiac tissue model. The bioink incorporates poly(octamethylene maleate (anhydride) citrate) (POMaC) for elasticity\, starch as a porogen to generate interconnected pores\, and carbon nanotubes (CNTs) to enhance electrical conductivity. Using extrusion-based 3D printing\, we fabricated a cone-shaped ventricular scaffold capable of supporting cardiomyocyte attachment and organization. Human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) seeded onto the scaffold exhibited spontaneous and synchronized contractions\, demonstrating the potential of this platform to support functional cardiac tissue formation. This 3D-printable model provides a promising platform for investigating cardiac tissue development\, disease modeling\, and future therapeutic applications.\nSupervisor Name: Milica Radisic\nYear of Study: 4\nProgram of Study: PhD\nPowered by Calendly.com

Recordings taken during DBS surgeries have identified abnormally elevated beta oscillations (13-30 Hz) in the basal ganglia thalamo-cortical (BGTC) circuit. Excessive beta oscillations are strongly correlated with the motor symptoms of PD\, such as bradykinesia and rigidity\, making them a critical target for therapeutic intervention. Research indicates that improved motor outcomes seen in patients following DBS treatment are the result of stimulation suppressing abnormal beta activity in the BGTC circuit. Recent studies from our lab indicate that DBS not only modulates beta oscillations but may affect changes at the molecular level. A major biomarker of PD is the aggregation of alpha-synuclein (a-syn) in the brain\, which has been implicated in dopaminergic (DA) cell death in the substantia nigra. DBS was shown to reduce toxic alpha-synuclein (a-syn) aggregation and DA neurodegeneration in preclinical models of PD. This suggests DBS engages a mechanistic relationship between aberrant circuit activity and the molecular pathology underlying neurodegeneration\, and that DBS suppression of beta oscillations may elicit disease-modifying changes in PD. In this study\, we hypothesize that excessive beta activity in the BGTC circuit worsens a-syn aggregation and DA neurodegeneration in PD. To address this question\, we will use a genetic rat model of PD previously validated in our lab. This model recapitulates a-syn aggregation\, DA neurodegeneration\, motor dysfunction\, and disease-associated beta activity. In this model\, we will chronically elevate beta oscillations using a technique called optogenetics. This method combines genetic engineering and light to activate or inhibit select neurons when exposed to specific wavelengths. We will use a 20 Hz pattern of light stimulation in basal ganglia structures\, which has been previously shown to elevate beta oscillations in the BGTC circuit. Electrophysiological methods will be used to validate emergence of elevated beta oscillations in the circuit during optogenetic stimulation. Behavioral tests used in our lab’s previous work will be applied to compare how elevated beta correlates to a PD motor phenotype. Immunohistochemistry will then be used to compare DA neurodegeneration and a-syn pathology in the experimental versus control conditions. If we find that the molecular pathology is worsened in the experimental condition\, future work will focus on modifying DBS strategies for maximal beta suppression\, including validation of whether this approach halts the progression of PD.\nSupervisor Name: Luka Milosevic\nYear of Study: 2\nProgram of Study: PhD\nPowered by Calendly.com
