DESCRIPTION:Speaker\nJaideep Bains\nDirector \nKrembil Research Institute\nUniversity Health Network \n\n\nPlease join us as Jaideep Bains\, Director of the Krembil research Institute\, shares his research with us.\nResearch Summary\nDr. Bains’ research uses multiple experimental approaches to reveal key features about neural circuits that regulate internal state and are also purposed to control behavior and store information in response to challenges. Specifically\, his lab uses in vitro slice electrophysiology\, in vivo imaging\, optogenetics\, behavioral analysis tools and physiological assays to characterize neural circuits that decode stress\, modify internal states and generate specific coping behaviors. One of his goals is to better understand the mechanisms that allow these circuits\, or specific cell populations\, to store information related to the modality\, intensity and temporal features of stress. In addition to his extensive experience in synaptic physiology and electrophysiology he has used circuit mapping approaches to link activity in specific cell populations to different behaviors. His work has linked brief stress exposure and enduring synaptic changes in the hypothalamus (reviewed in Bains et al\, Nat Rev Nsci\, 2015). His lab has provided clear evidence supporting a role for astrocytes in controlling the strength of excitatory synapses in the hypothalamus (Gordon et al\, Nat Nsci\, 2005\, Neuron\, 2009). More recently\, his lab has shown new roles for hypothalamic CRH neurons as bottom-up controllers for complex behaviors associated with stress coping (Fuzesi et al\, Nat Comm\, 2016)\, the transmission and detection of affective states between mice (Sterley et al\, Nat Nsci\, 2018) and linking stress controllability and active behaviour strategies (Daviu et al\, Nat Nsci\, 2020).

While studies have investigated the role of descending corticospinal commands and spinal reflexes in maintaining balance using methods such as motor evoked potentials (MEPs) using transcranial magnetic stimulation (TMS) and H-reflexes using peripheral nerve stimulation\, the exact pathways involved remain unclear. We aim to contribute to this understanding by exploring the effects of posture on spinal motoneurons by utilizing F-waves as a direct measure of their excitability. Furthermore\, we wish to further investigate corticospinal and spinal pathways by isolating the effects of body position and both voluntary and postural muscle activity to understand the specific factors influencing postural control.\nData collection from healthy participants is ongoing. Participants performed four tasks: natural standing\, supported standing with voluntary muscle contraction\, supported standing\, and sitting. Electromyographic signals from the soleus\, medial head of gastrocnemius\, and tibialis anterior muscles was recorded. Visual feedback was used to ensure consistent muscle activity during voluntary contraction to match natural standing. Soleus F-waves\, H-reflexes\, and MEPs were measured.\nResults from 16 participants (8F) indicate that MEP measures of cortical excitability show modulation of the silent period. A shorter silent period in voluntary activation indicates decreased cortical inhibition\, thus increased cortical excitability compared to standing. Meanwhile\, F-wave measurements of spinal excitability shows that spinal motoneuron excitability is not modulated by position or activity. This indicates that modulation due to voluntary activation does not originate in the spinal cord. These adverse events are mainly characterized as prolonged reduction of airflow and respiratory rate and reduced blood oxygen saturation. The consequences are cardiopulmonary complications\, increased morbidity\, hospital length of stay\, costs\, 30-day readmission\, and higher mortality. The risk factors include age\, sex\, body mass index\, opioids’ dosage\, comorbid sleep and cardiac disorders\, and concomitant use of other medications. While identifying high-risk individuals\, proper monitoring of their respiratory function\, and timely interventions can significantly prevent the adverse events\, current techniques have low accuracy\, lack external validation\, have false errors\, cannot predict adverse respiratory events. Therefore\, in this research project\, our goal is to develop algorithms based on signal processing and artificial intelligence techniques for risk assessment and prediction of respiratory adverse events during sleep in individuals who are taking opioids. The outcome of this research can help identify high-risk individuals for more frequent monitoring of their respiratory function. SUMMARY:Tunable Multifunctional Macromolecules via Parallel Experiment and Computation: from Drug Delivery to Nucleic Acid Therapeutics - Theresa Reineke
DESCRIPTION:Speaker\nTheresa Reineke\nProfessor \nDepartment of Chemistry\nCollege of Science & Engineering\nUniversity of Minnesota \n\nAbstract\nMultifunctional macromolecules have a tremendous impact on a multitude of applications. For example\, the development of polymer excipients can improve the solubility and bioavailability of intractable drugs\, nucleic acid delivery vehicles have promise to promote selective genome editing\, and materials from sustainable monomers may lower the environmental impact of plastics. While the materials function is diverse across these fields\, the synthetic principles are similar and should be readily tunable for each specific application while remaining biologically and environmentally benign. Modern polymer chemistry and engineering offers numerous pathways for tailorable materials development and a multitude of rich monomer functionality (high heteroatom content and stereochemistry)\, composition\, molecular weight\, and architecture (linear\, statistical\, block\, brush)\, where engineering is now hindered by the infinite design space. To this end\, we have coupled parallel experimental workflows with computation and/or machine learning to effectively understand structure-activity trends\, mechanisms\, and rapidly discover high performance structures. For example\, we have applied these techniques to design delivery vehicles for Cas9 ribonucleoprotein (RNP)\, plasmid (pDNA)\, and combination payloads. A library of statistical copolymers has been synthesized and systematic variations in physicochemical properties and trends for encapsulation/delivery of RNP and pDNA payloads were examined in parallel. Structure-function correlations underlying cellular internalization\, editing efficiency\, gene expression\, and cellular toxicity of the polymer-payload formulations were probed through machine learning approaches. This workflow allowed rapid identification of successful architectures that outperform commercial reagents\, achieving nearly 60% editing efficiency via non-homologous end-joining. Furthermore\, the data set allowed us to uncover the physicochemical basis of delivery performance\, offering understanding of divergent requirements for successful delivery of RNP and plasmid payloads. Our work demonstrates the power of combinatorial synthesis and high-throughput characterization methodologies coupled with data science approaches to discover promising macromolecules and structure-property relationships that would have otherwise remained inaccessible to intuition. \nBiography \nTheresa M. Reineke is the Prager Endowed Chair in Macromolecular Science and a Distinguished McKnight University Professor in the Department of Chemistry at the University of Minnesota. She also holds graduate faculty appointments in the Departments of Chemical Engineering/Materials Science and Pharmaceutics. She received a B.S. Degree from the University of Wisconsin-Eau Claire\, a M.S. Degree from Arizona State University\, and a Ph.D. from the University of Michigan. She then received a National Institutes of Health Postdoctoral Research Fellowship for her work in gene therapy at the California Institute of Technology prior to beginning her independent faculty career. Her research group is focused on enabling fundamental and applied technology advancements in the fields of macromolecules for nucleic acid delivery and gene editing\, oral delivery of therapeutics\, and sustainability. She has published over 200 peer-reviewed manuscripts and patents and manages a large group of researchers supported by several corporate\, private and national funding agencies. Reineke is a Fellow of the American Chemical Society\, Royal Society of Chemistry\, along with the Kavli and Alfred P. Sloan Foundations. She has received numerous awards\, including in the 2005 National Science Foundation CAREER and Beckman Foundation Young Investigator Awards\, 2008 Camille and Henry Dreyfus Teacher-Scholar Award\, 2009 National Institutes of Health Director’s New Innovator Award\, 2012 Outstanding New Investigator Award from the American Society of Gene and Cell Therapy\, 2017 Carl S. Marvel Creative Polymer Chemistry Award from the American Chemical Society Division of Polymer Chemistry\, 2018 DuPont Nutrition and Health Sciences Excellence Medal\, and 2022 Arthur C. Cope Scholar Award from the American Chemical Society. Reineke has also served for 11 years as an Associate Editor for the journals ACS MacroLetters and Chemical Science and in 2023 became Editor-in Chief of Bioconjugate Chemistry. Further\, Reineke has cofounded three biotech companies in the field of nucleic acid delivery: Techulon\, Inc.\, Nanite\, Inc.\, and LiberateBio\, Inc.

DESCRIPTION:Speaker\nGeoffrey Wright\nProfessor \nCollege of Public Health\nTemple University \n\nAbstract\nThis novel application of virtual reality (VR) head-mounted display (HMD) technology has been designed as a balance assessment device that establishes a method of assessing fall-risk by using a series of automated postural tests with different levels of somatosensory\, visual\, and visual-vestibular reliability. Our other automated VR-based tools for assessment of vestibular and oculomotor function further establish the sensorimotor integrity of these balance-related processes. We have established construct and criterion-measure validity against the research-grade laboratory measurement devices\, such as motion capture cameras and force-plates and the gold-standard in clinical posturography (Neurocom)\, and to commonly used clinical assessments\, such as the Berg Balance Test\, Tinetti test\, TUG\, VOMS\, vHIT\, and DVAT. The construct validity of our assessment approach has been validated in various populations such as veterans\, concussion\, vestibular issues\, PTSD\, veterans\, and older adults with fall risk. Accurate and expedient diagnosis significantly increases a clinician’s ability to plan the most effective treatment plan. Following assessment\, providing custom-designed rehabilitation tools that are gamified\, progressive\, automated\, and specifically targeted to identified deficits\, a patient’s treatment can be motivating\, quantified and recorded\, and more effective. In summary\, by creating portable\, easy-to-use\, accessible tools\, such solutions can be incorporated into the clinical care pipeline even in remote\, austere\, or underserved communities and facilities.
