SUMMARY:Graduate Student Seminar Series - Raymond Hawkins
DESCRIPTION:Graduate Student Seminar Series\nPlease ensure you invite your Principal Investigator by adding their email via the ‘Add Guest’ button and they will also be notified of your presentation.\nLocation: HS610 – 155 College St\, Room 610\nPresentation Title: ReSCU-Nets: Recurrent U-Nets for segmentation of timelapse microscopy data\nAbstract: Segmenting multi-dimensional microscopy data requires high accuracy across many images (e.g. timepoints or Z slices) and is thus a labour-intensive part of biological image processing pipelines. We present ReSCU-Nets\, recurrent convolutional neural networks that use the segmentation results from the previous frame as a prompt to segment the current frame. We demonstrate that ReSCU-Nets outperform state-of-the-art image segmentation models in different segmentation tasks on time-lapse microscopy sequences.\nSupervisor Name: Rodrigo Fernandez-Gonzalez\nYear of Study: 3\nProgram of Study: MASc\nPowered by Calendly.com

Most CVD cases can be prevented at an early stage through targeted interventions. Regular exercise is the most effective method to reduce these risk factors\, with studies showing that engaging in 150-300 minutes of moderate-intensity aerobic exercise or 75-150 minutes of vigorous-intensity exercise per week can significantly mitigate CVD risks. However\, individuals with pre-existing cardiovascular risk factors face heightened dangers when exercising in extreme environmental conditions\, such as in very cold temperature (below 10°C) or in very hot temperature (above 30°C). Exercising in these adverse temperatures can increase the likelihood of heat-related illnesses and cardiovascular strain\, leading to greater thermal and cardiovascular demands and heightened fluid needs. Despite these risks\, individuals may engage in physical activity to maintain a healthy life\, especially in real-world scenarios where temperature variations are unavoidable. Currently\, there is a lack of all-encompassing wearable technology to monitor and most importantly\, notify users of cardiovascular risk during exercise in adverse environments.\nSupervisor Name: Dr Shehroz Khan\nYear of Study: 2\nProgram of Study: MASc\nPowered by Calendly.com

This underscores the need for more equitable pulse oximetry strategies that account for\npigmentation to reduce this bias. Here\, we developed a wearable reflective pulse oximeter to gather raw\nphotoplethysmography signals. Conventional and modified pulse oximeter algorithms were applied based on the ratio-of-\nratio’s method and a melanin term\, derived from quantitative histology of the epidermis. This melanin-inclusive pulse\noximetry expression was developed and validated in a Hampshire porcine model (n=4) using adjacent areas of darkly and\nlightly pigmented skin. The animals were exposed to controlled hypoxia with saturation levels ranging from 100% to 70%.\nBlood gas analysis was performed and synchronized to the pulse oximeter readings on dark and light sections\, with\nepidermis biopsies sampled and Fontana-Masson stained for pigment quantification. The melanin-inclusive approach\nreduced the bias in both the 100-85% (from 1.85% to 0.62%) and 84-70% saturation ranges (from 2.70% to 0.16%). This\nwas a marked improvement over conventional pulse oximetry and reference medical-grade devices\, which exhibited higher\nbiases (3.08% to 4.98%). This study suggests that the bias in oximeter readings may be correlated with physical quantities\nof melanin in the epidermis. Future work will entail obtaining a melanin coefficient from the wearable device itself for a\ncomplete on-board non-invasive solution. This melanin-inclusive method is a promising strategy to reduce pulse oximeter\nbias and improve the accuracy of a widely-used medical device\, potentially enhancing care across diverse patient\npopulations.\nSupervisor Name: Daniel Franklin\nYear of Study: 4\nProgram of Study: PhD\nPowered by Calendly.com

Currently\, there are many new synthetic biology based engineered cells in the research and discovery phase with the hope of emerging as new therapeutics. We believe that once most of the studies are passed this phase\, there is a safety problem that prevents them from unleashing their full potential. Through my PhD projects\, we proposed a strategy for decoupling HLA binding from engineered proteins that we think might overcome the safety problem.\nSupervisor Name: Michael Garton\nYear of Study: 6\nProgram of Study: PhD\nPowered by Calendly.com
