The mechanisms of action of DBS on\nthe collective activity of neuronal circuits is not fully understood. We use a recurrently-connected\nexcitatory-inhbitory network based on the Brunel network architecture that can produce activity in\na variety of states. Using a model of DBS that can reproduce observed effects such as antidromic\nactivation\, local somatic suppression\, and axonal activation\, we characterize the effect of stimulation\nacross the entire parameter space of the network. We show that the effects of stimulation are de-\npendent on the baseline state of the network\, with the level of beta suppression dependent on the\nlevel of inhibition and the external drive. Specifically\, networks with higher inhibition and lower drive\nshow greater disruption of beta oscillations. Continuous monitoring of daily routines can further support the longitudinal assessment of functional ability and early signs of cognitive decline in older adults living independently. Despite significant progress\, ADL monitoring systems still face a trade-off between accuracy\, computational complexity\, and the need for continuous video recording.\nTo address this gap\, this thesis proposes approaches to enable accurate\, non-visual\, and lightweight multimodal ADL monitoring systems suitable for edge deployment. The contributions are organized around three interconnected research components.\nFirst\, we propose a novel lightweight fusion-based architecture\, called EFACT (Attention-Based Cross-Modal Temporal Modeling for Fine-Grained Activity Recognition Using RGB-D Data)\, designed for fine-grained human activity recognition using RGB and depth modalities. Using the Toyota Smarthome Dataset\, we demonstrate that EFACT outperforms state-of-the-art approaches under the established benchmark protocols for held-out participants and camera viewpoints. Additionally\, we show that the integration of quantization techniques (PTQ and QAT) allows EFACT to operate efficiently on resource-constrained platforms.\nThe second contribution of this thesis is the exploration of Non-Intrusive Load Monitoring (NILM)\, or estimating appliance-specific consumption from aggregate smart meter signals\, as a building block for non-visual ADL recognition. We propose EdgeTCNformer\, a lightweight causal framework for appliance-level power and state estimation that supports efficient local inference on residential edge gateways. Evaluation on four diverse datasets demonstrates a favorable accuracy–computational-efficiency trade-off under repeated sample-level and leave-one-house-out protocols\, while zero-shot cross-dataset evaluation exposes substantial limitations under regional and acquisition-domain shift.\nThe third contribution of this thesis is E2R-FuseNet\, an ADL recognition system for residential settings that combines NILM with millimeter-wave (mmWave) radar. This research aims to reduce those barriers by developing an AI-driven framework for real-time\, two-way sign language translation. The long-term goal is to combine Sign Language Recognition (SLR) and Sign Language Production (SLP) into a single system that enables more natural and accessible communication between Deaf and hearing communities.\nThe current stage of this research focuses on the SLR component using the American Sign Language Lexicon Video Dataset (ASLLVD). A complete preprocessing pipeline was developed to clean and organize the dataset\, extract body and hand landmarks using MediaPipe\, and convert sign language videos into machine-learning-ready representations. Three deep learning architectures\, LSTM-only\, CNN+LSTM\, and CNN+Transformer\, were then implemented and compared to identify the most effective approach for recognizing sign language from video.\nAmong the evaluated models\, the CNN+Transformer model achieved the strongest performance\, demonstrating a greater ability to capture both the spatial and temporal patterns that characterize sign language. These findings provide a strong foundation for the next phase of the project\, which will extend the model to generate natural\, expressive sign language from spoken or written language.\nThis work demonstrates how artificial intelligence can make communication more accessible across different languages and modalities. STIs are often asymptomatic and\, if left untreated\, can lead to long-term health consequences that disproportionately affect women\, including pelvic inflammatory disease and infertility. Regular screening is therefore essential\, but current diagnostics are poorly suited for routine use: antigen-based POCTs lack adequate sensitivity while nucleic acid amplification tests require sophisticated laboratory infrastructure. Even where resources are abundant\, uptake remains low due to stigma and inconvenience. Clustered regularly interspaced short palindromic repeats (CRISPR)-Cas diagnostics offer programmable\, low-cost\, rapid\, isothermal nucleic acid detection. CRISPR diagnostics use controllable DNA cleavage upon detection of the target pathogen to generate signal. Traditional CRISPR diagnostics are fluorescence-based\, but recently CRISPR-responsive materials have emerged as an alternative to enable their use at the point-of-care. Here\, we design a multiplexed point-of-care CRISPR-Cas diagnostic platform incorporated into a tampon for at-home STI screening in women to detect Chlamydia trachomatis (CT)\, Neisseria gonorrhoeae (NG)\, and Trichomonas vaginalis (TV). The assay combines isothermal recombinase polymerase amplification and CRISPR-Cas12a reactions to detect species-specific nucleic acid sequences. Our platform leverages a DNA hydrogel with an immobilized colourimetric dye that is released by activated Cas12a in the presence of the target pathogen\, enabling equipment-free colourimetric readout\, eliminating the need for laboratory infrastructure. The CRISPR-Cas assay designs for CT detection in a standard fluorometric assay have a 1 aM limit of detection within 1 hour. Transitioning these designs to a hydrogel-based dye-release assay\, we achieve 500 aM sensitivity within 2 hours. The hydrogels will be tested with NG and TV CRISPR-Cas assays for multiplexed detection. This work presents a series of unimodal and multimodal implants featuring multi-wavelength photoplethysmography (PPG) and bioimpedance (BIOZ) sensing capabilities. Although lipid nanoparticles (LNPs) are clinically validated for RNA delivery\, their efficacy often declines when encapsulating larger mRNA transcripts necessary for genome editing. In this work\, we present a large-cargo-informed lipid discovery strategy that incorporates mRNA size into the rational design and screening of LNPs. By synthesizing a library of over 300 unique ionizable lipid candidates and evaluating their performance using multi-kilobase reporter mRNAs\, we identified a class of large-cargo-optimized lipids (LC-lipids). The lead ionizable lipid\, LC-1\, enabled improved Cas9 gene knockout and adenine base editing across multiple tissue types and administration routes\, including the liver (intravenous)\, brain (intrathecal)\, and lung (intratracheal)\, consistently outperforming benchmark LNPs using LP-01 and ALC-0315 as the ionizable lipid.\nSupervisor Name: Dr. Bowen Li\nYear of Study: 4\nProgram of Study: PhD\nPowered by Calendly.com
